How To Achieve Muffled/Low/Deep Vocal Effect

The vocals are just shifted down in pitch. They definitely aren't muffled, however, you can use a lowpass filter on an EQ if you ever want to get a muffled sound.

Changing the pitch affects the tempo of the vocal track, it sounds like slow motion speaking
 
Depends on what you are using to change the pitch. Plugins that work in real-time don't throw it off time. I use FL and there are a few ways to change pitch. The method that doesn't throw audio off time just involves turning the "pitch" knob down an octave. If you happen to use the same DAW, it's the pictch knob located next to a "Mul" and "time"(not sure if this one is the correct name) knob.
