what I usually do is find a 2 - 4 bar loop from a song that i want to sample, cut that part from the original song, make it a wav file, then put it on a sampler on the step sequencer and then open up the piano roll for that sample. I find the desired pitch, then stretch the note for the 2 or 4 bars i determined, then loop it over and over until i can match the tempo up to where it loops seamlessly. then i could add drums or whatever. that's a very basic way of doing it. of course there's no particularly "right" way to go about doing it, and there's probably over a hundred other ways to do it. perhaps the best way, like everyone else said is to experiment to see which way works best for you.
