you have to route the sample to the fx section, there are three ways to do this:
1- in the easy edit page go to output then select FX 1 or the fx block you want to use
2- in the sample edit page go to output then select FX 1, etc you get the idea right ?
3- and the one not many people know of, you have two possible output settings for each sample, set the first one to stereo out and keep yhe volume @ 127, then set the second one (output 2) to FX 1 or the FX block you want output @ 127 go to the FX section and set the mix @ 100% wet (wet=+63) then go to the FX output level parameter and adjust as needed
by doing it this way you are not inserting fx like in options 1 or 2 you are creating a send and a return (in paralell) so you get a more flexible FX routing, with six effect blocks you can do a lot of cool processing in the A5000 without any external FX
BTW make sure the FX are not in bypass or you won't be able to hear them, make sure the mix is not too low or you won't hear any fx, if the fx are in through there is no fx in the signal path
